MGC ESTs

The initial step in the identification of a full-length ORF clone from an MGC cDNA library is 5' EST sequencing. (Originally, one 384 well plate per library was also submitted for 3' sequencing but this practice was soon discontinued.) All these sequences are deposited in GenBank and are found at:

The sequence of each 5' EST is analyzed, and if it meets selected criteria (described in the PNAS paper "Generation and Initial Analysis of more than 15,000 Full-Length Human and Mouse cDNA Sequences"), and if it is also unique, it is designated as a Candidate Clones. It is then returned to the MGC pipeline for full insert sequencing and reanalysis to confirm the presence of a complete open reading frame.
