Human embryonic stem cells (ESCs) and human induce pluripotent stem cells (iPSCs) can provide theoretically unlimited supplies of human cardiac myocytes(CMs) for research and potentially clinical applications. Furthermore, generation of iPSCs from genetically diverse samples will enable creation of libraries of iPSCs that will better reflect the genetic diversity of the population as well as generate disease-specific models. However, the promise of these unique cells is only starting to be realized as technical roadblocks are overcome and proof-of-principle studies are undertaken. For example, the differentiation of human pluripotent stem cells to cardiomyocytes has been highly inefficient, but here we will described a novel approach using extracellular matrix in combination with growth factors to produce in high yield and purity CMs. The presentation will also describe the use of human iPSCs to model Pompe Disease and Long QT syndrome. The final section will describe a unique cardiac progenitor cell population that we have isolated using the SRF-cardiac specific enhancer which may prove useful in cardiac repair.

He will discuss his lab's recent work in the area of reprogramming the hematopoietic lineage to pluripotency. The Zambidis lab studies the formation of pluripotent stem cells and hematopoietic, endothelial and cardiac differentiation with a focus on the potential therapeutic use of pluripotent stem cell-derived cells. Projects include studying the developmental biology of hematopoiesis as well as improving the differentiation of human embryonic stem cells and human induced pluripotent stem cells to hematopoietic lineages including erythrocytes and lymphocytes; improving the differentiation of human pluripotent stem cells to endothelial cells capable of forming the vasculature system; and improving the differentiation of human embryonic stem cells (hESC) and human induced pluripotent stem cells (hiPSC) to the cardiac lineage. The research team also studies the developmental biology of gastrulation and cardiogenesis in model organisms and explores potential applications of pluripotent stem cell-derived cardiac in tissue engineering, regenerative medicine, cardiotoxicity screening and novel drug discovery. | Elias Zambidis, MD, PhD |

Control of cell fate and its extracellular environment is critical for tissue regeneration and cell therapy. This talk will explore methods to enhance the engraftment of systemically infused stem cells through engineering the surface of cells to induce a robust rolling response. This approach is based on leukocytes ability to target tissues via specific interactions with the vascular endothelium. In addition, a novel strategy to engineer cells with an intracellular depot of phenotype altering agents will be described that can be used for programming cell fate via both intracrine-, paracrine-, and endocrine-like mechanisms. The talk will also examine the potential of cell surface sensors that can be used to detect signals within the cellular nano-environment with unprecedented spatial and temporal resolution and that should be useful for elucidating niche biology in vivo.
